Part A - Part 2 of Figure 17.2

Part B - Figure 17.2 DNA sequence.

Is there evidience that the design of the DNA sequence in step 2 took E. coli codon bias into account?  HINT: See Table 17.1.

Part C - Expression of human insulin in E. coli using plasmid piB1 in Figure 17.2.

Which medium would be best for producing high levels of human insulin using an E. colistrain that contains the plasmid piB1?

Medium containing glucose as the sole carbon source.
Medium containing some other carbon source besides glucose and lactose.
Medium containing some other carbon source besides glucose and lactose with X-gal added to it.
Medium containing some other carbon source besides glucose or lactose with IPTG added to it.

Part A:

Using knowledge of the genetic code to deduce the coding sequence of a gene based on the sequence of a polypeptide.

Part B:

Yes, the sequence shows evidence that a least one known bias in codon usasge was taken into when it was designed.

Part C:

Medium containing some other carbon source besides glucose or lactose with IPTG added to it.
